Nicotinamide mononucleotide (NMN) in elderly men motor function is reformed

~Super-aging societyIssue"Sarcopenia"Expectation for preventive effect~

Points of Utterance:

      • Take 250mg of nicotinamide mononucleotide (NMN, Note 1) orally per day in healthy elderly men It has been suggested that it may improve muscle strength and performance.

This is the first NMN clinical trial published in an elderly man . The results revealed the possibility of improvement of muscle strength by oral intake of NMN in elderly men.

NMN Oral intake is effective for preventing age-related muscle weakness such as sarcopenia (Note 2) and preventive medicine in super-aging society Expected to be a new strategy.

Overview:Department of Diabetes and Metabolic Medicine, Faculty of Medicine, The University of Tokyo Assistant Professor Masaki Igarashi, Dr. Yoshiko Nakagawa, Doctor Masaomi Miura, Professor Toshimasa Yamauchi Research Group used healthy elderly men as test subjects. Ndinucleotide (NAD+, Note 3) nicotinamide mononucleotide, a precursor

Effects of (NMN) on aging phenomena including muscle weakness when taken orally A randomized, placebo-controlled, double-blind, parallel-group controlled trial (Note 4) was conducted. When healthy elderly men take 250 mg NMN orally for 12 weeks, It was clarified that NAD+ and related metabolites blood concentration rises, and motor activity such as walking speed and grip strength improve. In addition, NMN oral intake showed a tendency to improve hearing . In the super-aging society that Japan is facing, sarcopenia prevention is considered a major issue. From the results of this study, NMN sarcopenia by oral intake prevention effect is expected, and it will contribute to extending healthy life expectancy in the future It is considered.

The results of this research were published in the British scientific journal "NPJ Aging" online edition (*) on May 1, Japan time. This research was conducted as a contract study by Mitsubishi Corporation Life Science Co., Ltd. .

*This paper was scheduled to be published on April 25, but has been postponed to May 1.

Presentation contents: Research background

For the onset of ageing, diabetes, cardiovascular disease, cancer, Alzheimer's disease, etc. age-related diseases

NAD+Intratissue concentration reduction are closely related. So far, NAD+ is a precursor

Ingesting NMN has often been shown to restore age-related NAD+ decline and prevent age-related diseases in animal studies. However, in humans, NMN on ingestion and aging effects is not well known is the current situation. Therefore, the research group planned a placebo-controlled, randomized, double-blind, parallel-group study to clarify the safety and efficacy of taking NMNs in older adults.

Research Profile

In this study, 42 males aged 65 years and older healthy elderly were compared to NMN intake group (250 mg/day) and placebo.

(Note 5) Randomly assigned to the ingestion group, NMN or placebo intake was performed for up to 12 weeks. This clinical trial was approved by the Ethics Committee of the Faculty of Medicine, the University of Tokyo, and will be conducted in 2019 at the University of Tokyo Hospital Phase 1 Unit. Did.

First, 6 weeks (NMN group n=21, placebo group n=21) or 12 weeks (NMN Group n=10, placebo group n=10) After ingestion, general blood calculation ( number of red blood cells, white blood cells, platelets, etc.), biochemical test items

Change, NAD+ change in whole blood, Muscle strength change (30 second chair standing test, walking speed, grip strength), Bioimpedance method (Note: We evaluated changes in body composition due to 6), fatty liver by CT, changes in visceral fat amount, changes in hearing ability, etc.

Results of the longest 12 weeks NMN oral intake showed no apparent adverse events, including blood test results. The group that took NMN orally was compared to the group that took placebo orally, and the group that took NAD+ and NMN NAD+ precursors Blood levels were effectively increased (Figure 1).

Next, in order to investigate the effect of NMN oral intake on healthy elderly men skeletal muscle mass, the primary endpoints were measured skeletal muscle mass index (Note 7) and muscle mass by site, and the NMN group and the placebo group Before, 6th, 12th week Mixed effects model with mean value (Note 8) mixed-effect model for repeated measures(MMRM, Note 9) It was evaluated using the statistical analysis method. As a result, even in any analysis, no significant difference was found in skeletal muscle mass changes.

On the other hand, in order to investigate motor function, walking speed, 30-second chair standing test, grip strength were evaluated, and analyzed using the same statistical method. The results showed significant improvements in walking speed and left handgrip strength test after taking NMN by mixed effects model or MMRM. These results do not affect skeletal muscle mass if taken orally with continued ✰✰, healthy elderly men It has been found to improve motor function. There was also a significant difference in walking speed between 6 and 12 weeks after each group mean value . In addition, after 6 weeks 30 seconds of chair standing up test, the placebo group and the NMN group A significant difference was observed between the two.

Of the soother endpoints evaluated in this clinical trial, there were no items that showed significant changes in the NMN intake group, but in the right hearing ability, even if they are not statistically significant, there was an improvement. A trend was noted.

Social significance and future schedule

In this study, healthy older males took 250 mg NMN per day for 12 weeks. It was confirmed that it is possible to continue to ingest it safely and that there is a significant increase in NAD+ and NAD+ related metabolites in the blood. The results showed that NMN improves muscle strength and performance. Therefore, continuous oral intake of NMN is recommended for sarcopenia

It is considered to be a very effective means of preventing muscle weakness associated with aging. In the future, as we welcome a super-aging society, the anti-aging effect of NMN oral medicine for the elderly will be reduced to healthy life expectancy extension / society as a whole It is expected to contribute to the improvement of productivity.

We look forward to working with you to develop cost effective products and long term relationships with mutual benefit.
